Cell Culture Drug Resistance Testing (Chemotherapy Sensitivity and Resistance Assays) refers to laboratory testing of a patient's own cancer cells with drugs that may be used to treat the patient's cancer. A group of lab tests known as human tumor assay systems (HTAS) can aid oncologists in deciding which chemotherapies work best in battling an individual patient's form of cancer. Additionally, these studies serve as a useful step towards the development of an in vitro chemosensitivity test for human melanoma, and provide an operational quantitative basis for further exploration of in vitro-directed therapy in metastatic by:
A drug sensitivity test prior to clinical treatment is necessary for individualized cancer therapy. A useful in vitro model that mimics the in vivo tumor microenvironment is required to select optimal anti-cancer agents. To promote the microfluidic technology moving from the laboratory to the clinic, we developed a microfluidic-based perivascular tumor model that makes it possible to assess. Patients with melanoma, lung cancer, and sarcoma dominated the group. The most active in vitro drug was correlated per clinical trial. Thirteen of 17 (76%) sensitive in vitro predictions and 51 of 54 (94%) resistant in vitro predictions were accurate. The assay in this study had a sensitivity of 81% and specificity of 93%.Cited by:

Human Tumour Drug Sensitivity Testing In Vitro: Techniques and Clinical ApplicationsAuthor: K C Calman. Predictive drug testing on human tumor cells in order to define the appropriate chemotherapy will remain imperative as long as the anticancer agents available are.

The plating efficiency of tumour cells obtained from spheroids was always greater than for the cells obtained from the dissociated tumour of origin and was in Cited by: Although a number of chemosensitivity tests have been proposed since the advent of modern anticancer chemotherapy, interest has been renewed by the possibility of cloning human tumor cells on agar plates, with a view to testing drug activity on cells with high pro liferation capacity.
INTRODUCTION In Hamburger and Salmon [1] introduced an assay for the clonogenic cells of human tumours which has since been widely exploited in an attempt to put cancer chemotherapy on a rational footing by allowing the oncologist to choose the drugs which he will use to treat an individual patient on the basis of in vitro chemosensitivity tests of that patient's tumour by: 2.
The technical limitations of the test are presented, and it is concluded that with the present status of the test, it is only suited as a preclinical test system.
It is suggested that in vitro sensitivity testing of drugs in a panel of cell lines established from patients with small cell carcinoma of the lung might help to identify drugs that could alter the clinical course of the by: 4.
A new method to test the sensitivity of tumour cells to chemotherapy is presented. Tumour cells were incubated in vitro on agar, and drug-induced cell cycle perturbation was monitored by flow. The US National Cancer Institute (NCI) 60 human tumour cell line anticancer drug screen (NCI60) was developed in the late s as an in vitro drug Cited by: In vitro cytotoxic drug sensitivity testing of human tumour xenografts grown as multicellular tumour spheroids.
In so doing, they combine the latest technological developments with strategic outlooks, such as novel cell and tissue systems for drug screening and testing, as well as emerging in vitro culture technologies. Equally importantly, the book does not.

The xenografts models (nude mice) are used for testing the tumorigenicity and metastatic ability of cancer cell lines. Summary. Background: Tumor cell drug sensitivity is an important determinant of chemotherapy measurement in vitro would aid in therapy individualization and new drug development.

A human tumor cloning assay was used to analyze drug sensitivity profiles for 99 pairs of tumor samples taken simultaneously from the same patient. These pairs included two areas of the same primary (12 pairs), primary tumor versus metastasis (29 pairs), and metastasis versus metastasis (45 Cited by: Drug sensitivity testing utilizing preclinical disease models such as cancer cell lines is an important and widely used tool for drug development.
Importantly, when combined with molecular data such as gene copy number variation or somatic coding mutations, associations between drug sensitivity and molecular data can be used to develop markers to guide patient therapies.

1. Determining the drug sensitivity of tumor cells, based on the results selecting the possibly most effective drugs and/or drug combinations for the therapy. The result of the in vitro drug sensitivity of cancer cells can be used as a prognostic factor.
2. Ranking cancer drugs. To test the device, the researchers implanted it in mice that had been grafted with human prostate, breast, and melanoma tumors. These tumors are known to have varying sensitivity to different cancer drugs, and the MIT team’s results corresponded to those previously seen differences.Cell culture-based in vitro drug sensitivity tests may be able to predict the sensitivity of cancer cells to various molecular target drugs under development for future clinical application.
